ABSTRAK
Penyakit flu burung/ avian influenza merupakan kebobrokan infeksius atas ayam yang disebabkan oleh
virus pilek corak A yang teperlus di keluarga Orthomyxoviridae. Virus tersebut mempunyai rentang
inang yang luas, ialah beragam spesies burung, mamalia dengan manusia. Dewasa ini kaum peneliti
melaporkan terjadinya kenaikan jangkitan mikrob flu burung subtipe H5N1 atas mamalia dengan manusia.
Kondisi tersebut telah menimbulkan ancar-ancar bahwa mikrob flu burung yang bersirkulasi di lapangan mungkin
mengalami mutasi masam amino yang bertanggung balasan di pembelengguan reseptor yang disebut sebagai
tapak perlekatan reseptor (TPR). Penelitian ini dirancang untuk melancarkan karakterisasi molekuler
pada adegan gen HA yang diperoleh TPR atas mikrob flu burung subtipe H5N1 yang diisolasi dari berbagai
spesies sejak tahun 2003 cukup 2008. Amplifikasi adegan gen HA dilakukan dengan reverse transcriptase
polymerase chain reaction (RT-PCR). Semua produk RT-PCR positif selanjutnya disekuensing untuk
mengetahui susunan nukleotida adegan gen target. Hasil sekuensing tersebut akan datang dianalisis dengan
program Mega 4.0, yang meliputi: multiple alignment, deductive amino acid prediction, dan phylogenetic
tree. Hasil investigasi membuktikan kaum masam amino TPR yang bersifat abadi dengan diindikasikan
cenderung untuk bertalian dengan reseptor corak unggas, ialah masam sialat α 2, 3 galaktosa. Satu isolat
virus flu burung yang diteliti, ialah A/ layer/Jabar/MHW-RBS-02/2008 cecap delesi masam amino
posisi 129 dengan mutasi masam amino kedudukan 151 isoleusin jadi treonin. Analisis tanaman kekerabatan
menunjukkan pengelompokan mikrob flu burung tersebut tak membuktikan cermin distribusi tertentu dari
spesies ungas mau juga ceceran geografis akar mikrob yang diteliti.

ABSTRACT
Avian Influenza (AI) is an infectious disease in poultry, caused by type A of avian pilek virus
(AIV), in the family of Orthomyxoviridae. Almost all birds’ species are sensitive to the AI. Beside the
ability to infect various species of poultry. AIV type A has a wide range of host including all bird species,
mammals, dengan human. Today some scientists reported that the cases of AI in mammals, including humans
are increasing. This condition suggests that the AI mikrob circulated in the field may have some mutations
in the amino acid determinants responsible receptor binding site (RBS). A research was therefore designed
to investigate the molecular kelas of HA gen fragment responsible for receptor binding site of AIV isolated
from various poultry since 2003 to 2008. Molecular characterization was based on the amplification of
receptor binding site of HA gene by reverse transcriptase polymerase chain reaction (RT-PCR). All RT-
PCR of HA gene positive products were sequenced to determine the nucleotide composition at the targeted
fragment. Sequences yielded were analyzed by acara Mega 4.0 versions, including multiple alignment,
deductive amino acid prediction, and establishment of phylogenetic tree. The results show that all AIV
isolates could be determined of some conserved amino acids residues responsible for RBS which indicate
the binding preference of avian like receptor, sialic acid α 2, 3 galactose except isolate A/Layer/Jabar/
MHW-RBS-02/2008 which could be found a deletion of amino acid at position of 129 dengan mutation of 151
isoleucine into threonine. Phylogenetic study showed that clustering of AIV did not base on species of bird
or geographic origin of AI viruses which were studied.

PENDAHULUAN
Penyakit flu burung/ avian influenza (AI)
di Indonesia, atas awalnya kedapatan sebagai
penyebab kematian ayam yang layak tinggi
di sebanyak peternakan unggas petelur komersial
di Jawa Barat dengan Jawa Tengah, sejak tahun
2003. Penyebaran kebobrokan tersebut terus
meluas ke beragam propinsi lain, dengan pada
tahun 2007 hampir sarwa propinsi dilaporkan
endemis kebobrokan AI atas unggas, kecuali
Propinsi Maluku dengan Gorontalo (Komnas FBPI,
2007). Dewasa ini perkembangan jangkitan virus
flu burung H5N1 dinilai mulai mengkuatirkan,
karena becus melewati sawar/ barrier
pertahanan antar spesies. Beberapa laporan
yang mendukung data tersebut di atas, antara
lain afair kebobrokan AI subtipe H5N1 atas dua
harimau (Panthera tigris) dengan duet leopards
(Panthera pdanus) atas sebuah kebun binatang
di Thailand (Kheawacharoen et al., 2004), pada
kucing dengan anjing (Songserm et al., 2006-a dan
Songserm et al., 2006-b ), atas babi (Takano et
al., 2009), dengan atas bani Adam yang mula-mula kali
dilaporkan atas bulan Mei 1997 di Hongkong
(Subbarrao et al., 1998).
Kemampuan mikrob flu burung untuk dapat
menginfeksi pembantu ditentukan oleh hemaglutinin
(H/HA), ialah spike glikoprotein terluar yang
berbentuk suatu homotrimer dengan dikode oleh
segmen gen ke-4 dengan panjang 1778 nt. (Cox
dan Kawaoka, 1998). Monomer H/HA terdiri
dari bagian kaput yang berupa globulus yang
dihubungkan dengan fibrous stalk domain dan
terdiri dari duet irisan polipeptida, ialah H1
dan H2. Kedua irisan tersebut terbentuk
dengan cara jalan lepas (cleavage) oleh enzim
inang di alun-alun cleavage site (CS) (Horimoto dan
Kawaoka, 1997; Horimoto dengan Kawaoka, 2001;
Horimoto dengan Kawaoka, 2005).
Segmen H1 juga mengdanung semua
pendana fiil antigenik yang bersifat protektif
dan receptor binding cavity (RBC). Segmen
tersebut sebagai kampil yang terletak pada
ujung distal HA1 yang membentuk tempat
untuk melilit masam sialat dengan menjadi
perantara untuk perlekatan HA dengan
permukaan sel inang, (Roger et al., 1983, sitasi
oleh Naeve et al., 1984). Dalam RBC tersebut
terdapat masam amino yang bertanggung jawab
pada receptor binding site (RBS) alias tapak
perlekatan reseptor (TPR) (Cox dengan Kawaoka,
1998; Kovacova et al., 2002). Asam amino pada
TPR berperan bena untuk bertalian dengan
asam sialat atas ujung terminal oligosakharida,
antara beda α 2,6 sialyl lactosa, N-
acetylneuraminic acid α 2,6 Galactosa dengan α
2,3 sialyllactosa, Neuraminic acid α 2,3
Galactosa (Suzuki et al., 2000). Pada umumnya
semua struktur HA mikrob flu burung
mempunyai konfigurasi RBC yang mirip, terdiri
dari tiga elemen struktural, ialah α-heliks ( 190-
heliks, HA1 : 188 cukup 190), dengan duet loops,
yaitu 130-loops ( HA1:134 cukup 138) serta
220-loops (HA1: 221 cukup 228). Pemberian
nomor tersebut berdasarkan sistem penomeran
virus influenza subtipe H3. Menurut Zhou et
al., (2007) kaum masam amino bena yang
bertanggung balasan atas TPR dengan bersifat
lestari atas mikrob flu burung subtipe H5N1,
yaitu atas kedudukan Y 91, W 149, I 151, H 179, N
182, E 186, 190 L dengan 192 Q (H5 numbering
system). Residu masam amino atas RBC tersebut
merupakan conserved residu yang penting
dalam spesifitas pembelengguan reseptor atas virus
flu burung subtipe H1 dengan H5 (Kovacova et al.,
2002; Steven et al., 2006; Zhou et al, 2007),
disamping masam amino tersebut, ampas posisi
191 (Kovacova et al.,2002), kedudukan 132 (Muramoto
et al., 2006), dengan kedudukan 129 (Smith et al., 2006)
juga merupakan masam amino bena atas TPR.
Asam amino tersebut atas umumnya bersifat
lestari di spesifitas pembelengguan reseptor
virus influenza subtipe H1 dengan H5 (Kovacova et
al., 2002; Steven et al., 2006; Zhou et al. 2007).
Proses jangkitan mikrob flu burung, tidak
semata-mata ditentukan oleh patogenisitas
virus flu burung saja, melainkan juga oleh
kemampuan mikrob tersebut untuk berikatan
dengan reseptor inang. Untuk itu diperlukan
kesesuaian TPR yang sedia atas mikrob flu burung
dengan reseptor atas latar sel inang.
Sejauh ini kedapatan bahwa masam sialat (SA) α
2,3 galaktosa merupakan reseptor untuk virus
influenza akar ayam dengan hewan, sedangkan
reseptor α 2,6 galaktosa merupakan reseptor
untuk mikrob influenza akar manusia. Naeve et
al., (1984) dengan Gambaryan et al., (2006)
melaporkan bahwa kemampuan mikrob influenza
dalam menembus barikade penjagaan spesies,
diduga erat berhubungan dengan pergeseran
preferensi mikrob tersebut untuk berikatan
dengan reseptor SA α 2,3 galaktosa ke AS α 2, 6
galaktosa alias sebaliknya.
Kasus kebobrokan flu burung atas unggas
masih berlaku menurut kadang-kala di beberapa
wilayah di Indonesia. Dewasa ini telah
dilaporkan banyak afair kebobrokan flu burung
yang berlaku atas mamalia teperlus manusia.
Kondisi tersebut menimbulkan ancar-ancar bahwa

peningkatan afair jangkitan mikrob flu burung pada
mamalia dengan bani Adam tersebut mungkin
berhubungan dengan metamorfosis lingkungan penting
pada TPR mikrob flu burung akar unggas. Untuk
menjawab persoalan tersebut diperlukan
penelitian atas tingkat molekuler khususnya
pada adegan gen HA yang diperoleh TPR, pada
virus flu burung subtipe H5N1 akar berbagai
unggas yang diisolasi sejak tahun 2003 sampai
2008.
METODE PENELITIAN
Virus yang diteliti
Materi elementer investigasi ini berupa isolat
virus flu burung akar kaum spesies unggas
yang dikoleksi sejak tahun 2003 cukup 2008
dari Jawa Tengah, Daerah Istimewa
Yogyakarta, Jawa Barat, Jawa Timur, dan
Lampung. Isolat tersebut telah diidentifikasi
secara serologis dengan molekuler sebagai mikrob flu
burung subtipe H5N1 oleh penulis. Isolasi,
propagasi, dengan teknik molekuler dikerjakan di
Laboratorium Mikrobiologi, Fakultas
Kedokteran Hewan, UGM dengan fasilitas
biosafety cabinet kelas III. Sekuensing dilakukan
di Laboratorium Biotechnology, (PT. Charoen
Pockphan Indonesia, Jakarta).
Perbanyakan Virus.
Propagasi isolat mikrob flu burung dilakukan
secara in ovo menggunakan telur ayam
berembrio spesific pathogen free (SPF) umur 11
hari. Adanya pertumbuhan mikrob flu burung
ditentukan dengan tes hemaglutinasi (HA) dan
dilanjutkan dengan tes batu ganjalan hemaglutinasi
(hemagglutination imhibition/HI) mengguna-
kan anti serum distingtif mikrob flu burung subtipe
H5N1. Hasil propagasi menurut in ovo tersebut
kemudian diperiksa menurut molekuler. Prosedur
isolasi dengan pengenalan tersebut membentuk pada
manual on animal pilek diagnosis dan
surveillance (WHO, 2002).
Isolasi RNA
Isolasi RNA mikrob AI dilakukan dengan
micro-to Midi RNA isolation kit (Invitrogen,
USA) bertemu dengan garis haluan stdanar yang
disarankan oleh Invitrogen. Pada prinsipnya,
sejumlah 0,2 ml isolat mikrob flu burung dalam
cairan korioalantois ditambahkan ke di 0,2
ml larutan pelisis yang mengdanung 0,002 ml
â-mercaptoethanol. Campuran tersebut,
kemudian disentrifugasi atas 12.000 x g selama
2 menit master 25ºC (refrigerated microcen-
trifuge, acuan 5804R, Eppendorf, Hamburg,
Germany). Supernatan dipindahkan ke dalam
tabung bersih dengan ditambah dengan 0,2 ml
etanol absolut. Sampel akan datang dimasukkan
kedalam RNA spin cartridge, dengan disentrifugasi
pada 12.000 x g selama 15 menit 25ºC. Cartridge
dicuci 1 x dengan 0,7 ml wash buffer I dengan 1 x
dengan 0,5 wash buffer II. Cartridge tersebut
kemudian dipindahkan ke di RNA recovery
tube. Sejumlah RNA diperoleh dengan
melakukan elusi cartridge tersebut dengan 0,03
ml RNAse-free water dengan cara disentrifugasi
pada kecepatan 12.000 x g selama 2 menit 25ºC.
Suspensi RNA yang diperoleh, akan datang dipakai
untuk template pada anggapan reverse
transcriptase-polymerase chain reaction (RT-
PCR).
Amplifikasi Gen HA yang Terdapat TPR.
Amplifikasi adegan gen HA yang terdapat
TPR dengan primer Lee et al., (2001) RBS
Forward: -5’ aca cat gcy car gac ata ct-3’- dan

target amplifikasi kedudukan 155 cukup 699, dan
produk produk amplifikasi sekitar 545 bp. Reaksi
dilakukan dengan kit Superscript III-one-step-
RT-PCR with platina Taq (Invitrogen)
menggunakan Gene-Amp PCR System 2400
(Applied Biosystem, USA). Master mix

amplifikasi merupakan sebagai berikut: 2x reaction
mix 25 μl, primer forward 2 μl, primer reverse
1 μl (konsentrasi primer 40 pM/μl), Superscript
III/RT-Platinum Taq Mix 2 μl, RNA-se free
water 10 ìl dengan template RNA 10 μl, sehingga
total bagian anggapan merupakan 50 μl. Siklus
amplifikasi tersebut adalah: RT reaction 500 C
selama 30 menit, Initial hot start 940C selama
3 menit, denaturasi 940 C selama 30 detik,
annealing 43,50C selama 1 menit, extension 680
C selama 90 detik dengan final extension 680 C
selama 4 menit. Reaksi tersebut dilakukan
sebanyak 40 siklus.
Produk PCR divisualisasi dengan metode
elektroforesis menggunakan gel agarose 2% dan
pewarnaan dengan ethidium bromide (0,5 μg/
ml). Pita DNA yang teramplifikasi diamati di
dalam ruangan gelap menggunakan UV
transluminator, untuk selanjutnya didokumen-
tasi.
Sekuensing dengan Analisis Hasil Sekuens
Secara prinsip sekuensing dilakukan
sebagai berikut: komoditas PCR dimurnikan
terlebih dahulu dengan menggunakan kit
purifikasi Exo SAP-IT (USB Corporation)
sebelum di buat sekuensing, bertemu metode
yang direkomendasikan oleh produsen. Master
mix terdiri atas: Big Dye Terminator Cycler
Sequencing kit 8 μL, primer μL (10 μM), dan
DNA template disesuaikan dengan panjang yang
mencapai 20 ng/kb, dengan bagian akhir mencapai
20 μl. Siklus sekuensing dilakukan dengan
mesin PCR yang meliputi pre-denaturasi pada
suhu 960C selama 5 menit, dengan peredaran PCR
selama 25 kali dengan program: denaturasi pada
suhu 960 C selama 30 detik, annealing 43,50 C
selama 30 detik, dengan perluasan atas master 600 C
selama 1 menit. Hasil peredaran sekuensing
selanjutnya dimurnikan dengan Big Dye
XTerminator Purification kit, bertemu protokol
produsen. Produk yang telah murni tersebut
selanjutnya dirunning dengan alat DNA
sequencer.
Hasil sekuensing ialah berupa urutan
nukleotida adegan gen HA alamat dianalisis
dengan perangkat bioinformatik acara Mega
4.0 yang meliputi: multiple alignment, deductive
amino acid prediction, pair wise distance
calculation dengan tanaman kekerabatan (Tamura et
al., 2007).
HASIL DAN PEMBAHASAN
Hasil amplifikasi adegan TPR dengan
desain primer yang disusun oleh Lee et al., (2001)
sesuai alamat amplifikasi ialah dengan produk
akhir 545 bp. Pita DNA yang teramati cukup
baik, tanpa teramati pita DNA yang bersifat non
spesifik. Primer tersebut juga dipakai oleh
peneliti Indonesia lainnya, seperti: Dharmayanti
et al., (2006); Dharmayanti dengan Darminto,
(2009), dengan produk layak baik. Contoh hasil
amplifikasi di investigasi ini disajikan pada

Hasil perbandingan masam amino penting
domain TPR tersebut dapat diamati atas Tabel
1. Menurut Zhou et al., (2007) kaum domain
penting yang bertanggung balasan atas tapak
perlekatan reseptor dengan bersifat abadi pada
virus flu burung subtipe H5N1, terletak pada
posisi Y 91, W 149, I 151, H 179, N 182, E 186,
190 L dengan 192 Q. Konsensus sekuens untuk sisi
kiri TPR merupakan kedudukan masam amino ke 130-134,
yaitu GVSSA dengan untuk bidang kiri kedudukan 220-
224 merupakan NGQSG (Kovacova et al., 2002; Zhou
et al., 2007). Kovacova et al., (2002) melaporkan
bahwa di antara masam amino tersebut, tujuh di
antaranya berhubung melantas dengan sel
reseptor inang, ialah Y 91, W 150, T 152, H
180, E 187, L 191, dengan Y 192 (H1 numbering
system). Posisi tersebut bertemu dengan H5 num-
bering system atas masam amino Y 91, W 149, I
151, H 179, E 186, L 190, dengan Y 191. Posisi asam
amino 152 T (H1 numbering system) tersebut
merupakan masam amino yang atas umumnya
ditemukan atas VAI subtipe H1, H2 dengan H3.
Posisi tersebut bagi Kovacova et al., (2002)
adalah concerved residue yang bersifat rendah,
dan memungkinkan penukaran masam amino
yang bersifat hidrofobik, misalnya: valin, leusin
dan isoleusin.

Hasil senada disampaikan oleh Muramoto
et al., (2006) yang melancarkan pengurangan mikrob AI
subtipe H5N1 isolat Vietnam atas tahun 2004
sampai 2005, membuktikan bahwa masam amino
penting atas TPR bertemu dengan laporan
penelitian Kovacova et al., (2002) dengan Zhou et
al., (2007), biarpun Muramoto et al., (2006)
menyatakan kecuali tujuh masam amino tersebut,
residu kedudukan 136 serin dengan 186 asparagin (H3
numbering system) bertemu dengan S 132 dan
N 182 (H5 numbering system) yang berperan
penting atas TPR. Menurut Smith et al., (2006)
residu bena di TPR yang belum
dilaporkan oleh peneliti lebih dahulu adalah
posisi 129 (H5 numbering system) yang pada
umumnya ditempati oleh serin.
Asam amino kedudukan 91 merupakan Y dan
menurut produk pengurangan terbukti kedudukan tersebut
bersifat abadi yang teramati atas virus
ancestral dengan mikrob flu burung yang diteliti sejak
tahun 2003 cukup 2008. Menurut Kovacova et
al., (2002) masam amino 91 Y merupakan asam
amino yang bersifat lestari, bahkan di antara
15 subtipe mikrob flu burung yang ada. Al-Majhdi
(2007) melaporkan bahwa kedudukan masam amino Y
91, bersama sama dengan masam amino kedudukan H
179, W 149, dengan I 151 menempati kedudukan pada
permukaan sialic acid binding site. Studi
dengan kristalografi membuktikan bahwa asam
amino tersebut berhubung dengan masam sialat
melalui belembang hidrogen.
Asam amino kedudukan 129 atas sarwa virus
flu burung di investigasi ini merupakan serin (S),
kecuali mikrob A/Layer/Jabar/MHW-RBS-02/2008
yang membuktikan delesi masam amino posisi
129. Secara am produk tersebut bertemu dengan
laporan Wu et al., (2008), ialah menurut umum
virus akar Indonesia yang teperlus Group A,
B, dengan C merupakan serin. Kondisi tersebut berbeda
untuk mikrob yang diperoleh dari gene bank, yaitu
A/Duck/Hunan/2005 dengan duet mikrob Indonesia,
yaitu A/Chicken/Jembrana/2004 dengan A/Human/
CDC/Indo/2006; kedudukan tersebut diisi oleh leusin.
Menurut Smith et al., (2006) kaum virus
yang diisolasi atas tahun 2004 cukup 2005,
menunjukkan mutasi S 129 L yang teramati
pada mikrob akar Vietnam dengan Indonesia. Virus
flu burung yang cecap mutasi S 129 L di
Indonesia dilaporkan teramati atas mikrob flu
burung dari Group C, ialah mikrob yang diisolasi
di Sumatera Utara atas tahun 2005. Substitusi
L 129 V dengan A 134 V, juga terdapat atas virus
flu burung isolat Thailand Th 676 DQ 360835
(Auewarakul et al., 2007). Substitusi tersebut
cukup menarik atas kedudukan keduanya berada
dalam loops 130 receptor binding cavity. Studi
pola hemaglutinasi yang dilakukan dengan virus
mutan L 129 V dengan A 134 V menunjukkan
peningkatan alternatif AS α 2,6 Gal., namun
jika mutasi tersebut hanya dilakukan atas L
129 V, mikrob mutan tersebut tak menunjukkan
perubahan alternatif reseptor.
Asam amino kedudukan 130, 131, 132, 133, dan
134 atas sarwa mikrob flu burung yang diamati
dalam investigasi ini bersifat lestari, yaitu
GVSSA. Menurut Kovacova et al., (2002) posisi
tersebut melenggekkan bidang kiri receptor binding
cavity dengan atas umumnya bersifat lestari,
meskipun untuk mikrob flu burung dengan subtipe
yang berlainan mempunyai susunan masam amino
yang berlainan pula. Sebagai contoh merupakan virus
influenza subtipe H1 kedudukan 130 cukup 134
tersebut relevan dengan kedudukan 131 cukup 135
(H1 numbering system) mempunyai konsesus
sekuen GVTAA. Posisi tersebut juga relevan
dengan masam amino nomer 134 cukup 138 (H3
numbering system) dengan sekuen GGSNA.
Mutasi masam amino kedudukan A 134 V bersama
dengan masam amino kedudukan L 129 V mempunyai
peran di menentukan alternatif reseptor
inang (Auewarakul et al., 2007).
Asam amino kedudukan 149 W atas mikrob flu
burung di investigasi ini teramati lestari.
Hasil tersebut bertemu dengan berita penelitian
terdahulu yang disampaikan oleh Kovacova et
al., (2002); Muramoto et al., (2006); Zhou et al.,
(2007), bahwa sejauh ini masam amino posisi
tersebut acap bersifat lestari. Menurut Al-
Majhdi (2007) triptofan (W) kedudukan 153 (H3
numbering system) bertemu dengan W 149 (H5
numbering system) bersifat non-polar yang akan
berinteraksi dengan masam sialat atas daerah
yang bersifat hidrofobik dengan belembang van der
Waals.
Asam amino kedudukan 151 merupakan isoleusin (I).
Diantara mikrob flu burung yang diteliti terdapat
satu isolat, ialah A/Layer/Jabar/MHW-RBS-02/
2008 yang cecap mutasi nukleotida
penyusun isoleusin kedudukan 151, ialah CTT menjadi
CCT sehingga menyebabklan mutasi asam
amino isoleusin 151 jadi threonine (T). Data
pesejajaran di antara subtipe mikrob influenza
yang dilakukan oleh Kovacova et al., (2002)
menunjukkan bahwa masam amino T 151 pada
umumnya dimiliki oleh mikrob influenza A
subtipe H1, H2, alias H3. Posisi tersebut bersifat
kurang stabil dibdaningkan ampas masam amino
lain atas lingkungan TPR dengan dapat bervariasi di
antara subtipe mikrob flu burung yang ada.
Menurut Zhou et al., (2007) masam amino
posisi 154, 155, dengan 156 merupakan tempat
potensial terjadinya glikosilasi. Secara umum
potensi glikosilasi atas isolat mikrob flu burung
yang diisolasi dari beragam spesies ayam pada

sejak tahun 2003 cukup 2008 dengan variasi
gejala klinis dengan patologis serta yang diisolasi
dari ayam dengan atau tanpa vaksinasi
mempunyai corak NST atas kedudukan 154, 155, dan
156. Menurut Shakin-Eshlemen et al., (1996) N-
linked glikosilasi atas umumnya berlaku pada
sekuens dengan corak N-X-S alias N-X-T. Posisi
X merupakan masam amino kecuali prolin. Kasturi
et al., (1995) melaporkan bahwa atas umumnya
sekuens N-X-T merupakan corak yang lebih
efisien untuk cecap penambahan rantai
samping karbohidrat (glikosilasi) dibandingkan
sekuens N-X-S. Glikosilasi atas lingkungan TPR
juga berlaku atas mikrob pilek subtipe H3,
terutama yang diisolasi selama dengan setelah
tahun 1975 (Schulze, 1997). Glikosilasi terjadi
pada masam amino kedudukan 126, yang merupakan
residu yang berdekatan dengan TPR dan
dilaporkan sebagai denai yang bertanggung
jawab atas fiil antigenik mikrob tersebut.
Iwatsuki-Horimoto et al., (2004) melakukan
analisis sekuens HA mikrob A/Ty/Ont/66 (H5N9),
dan mendapatkan duet titik potensial glikosilasi
pada kedudukan 131 dengan 158 (H3 numbering system),
namun atas mikrob flu burung HK/97 tidak
terdapat glikosilasi. Analisis bertambah berumur menun-
jukkan bahwa perbedaan glikosilasi pada
domain TPR tersebut tak dapat menjelaskan
perbedaan introduksi reseptor, biarpun kebe-
radaan karbohidrat atas kedudukan dekat domain
TPR, dapat meningkatkan afinitas dengan spesifitas
ikatan dengan reseptor. Menurut WHO (2005)
perubahan struktural HA dapat berlaku karena
mutasi alanin kedudukan 156 jadi treonin, sehing-
ga berlaku glikosilasi. Proses tersebut diprediksi
menurunkan afinitas terhadap reseptor.
Asam amino kedudukan 182 merupakan asparagin
(N). Sejauh ini kedudukan tersebut bersifat sangat
lestari baik atas masam amino maupun
nukleotida penyusunnya. Hasil yang sama
dilaporkan oleh Kovacova et al., (2002);
Muramoto et al., (2006); Zhou et al., (2007).
Asam amino atas kedudukan N 182 tersebut
menunjukkan ampas yang bersifat abadi di
antara subtipe HA yang beda (Kovacova et al.,
2002). Menurut Yamada et al., (2006) asam
amino kedudukan N 182 dengan glutamin (Q) 192
merupakan masam amino yang berperan untuk
menjaga kemantapan hubungan dengan reseptor
asam sialat. Mutasi ampas N 182 jadi L
atau Q 192 jadi R dianggap mampu
menyebabkan metamorfosis spesifitas reseptor SA
α 2,3 Gal., ke alternatif reseptor masam sialat α
2, 6 Gal. Secara alami mikrob yang mengalami
mutasi atas kedudukan tersebut dilaporkan terjadi
pada mikrob flu burung clade-2 yang diisolasi dari
orang di Azerbaijan dengan Irak.
Struktur α-heliks yang terdiri arah asam
amino kedudukan 186 glutamat (E), 190 leusin (L),
dan 191 tirosin (Y) bersifat concerved (Kovacova
et al., 2002; Al-Majhdi 2007; Zhou et al., 2007),
bahkan di antara subtipe HA mikrob flu burung
yang sedia (Kovacova et al., 2002). Asam amino
dan nukleotida kedudukan 192 tak menunjukkan
adanya mutasi, baik isolat mikrob flu burung awal
wabah cukup isolat tahun 2008. Menurut
Yamada et al., (2006) masam amino kedudukan N 182,
dan Q 192 berperan untuk melindungi stabilitas
ikatan dengan reseptor masam sialat inang.
Asam amino kedudukan loops 220 yang terdiri
dari kedudukan 220, 221,223, dengan 224 merupakan
receptor binding cavity bidang sebelah kiri (Kovacova
et al., 2002). Beberapa peneliti melaporkan
bahwa atas kedudukan tersebut, untuk mikrob flu
burung subtipe H5N1 mempunyai motif
NGQSG (Kovacova et al., 2002; Zhou et al.,
2007). Posisi tersebut berlainan untuk virus
influenza subtipe H1, ialah mempunyai motif
RGQAGR, sedangkan mikrob subtipe H3,
mempunyai corak RGQSGR (Kovacova et al.,
2002). Menurut Naeve et al., (1984) atas RBC
tersebut diperoleh ampas masam amino penting
yang berperan di alternatif pengikatan
reseptor atas mikrob influenza manusia, yaitu
asam amino kedudukan 226 dengan 228 (H2 dengan H3).
Posisi tersebut bersesuaian dengan masam amino
nomor 222 dengan 224 atas mikrob H5. Mutasi asam
amino kedudukan Gln 222 Leu dengan Gly 224 Ser
berperan meningkatkan kecondongan pengi-
katan reseptor corak ayam mengarah ke resep-
tor corak manusia, namun demikian mutasi
tersebut belum pernah dilaporkan atas virus
flu burung subtipe H5N1 (Gutierrez et al., 2009).
Menurut Yamada et al., (2006) mutasi asam
amino Asn 182 Lys dengan GLn 192 Arg atas virus
flu burung subtipe H5N1 menurut independen
mampu melantarkan metamorfosis pengikatan
preferensi reseptor corak ayam SA α 2,3 Gal.,
ke corak bani Adam SA α 2,6 Gal. Auewarakul et
al., (2007) melaporkan mikrob flu burung subtipe
H5N1 yang mempunyai preferansi reseptor SA
α 2,3 Gal., cecap metamorfosis preferensi
reseptor yang mengarah ke reseptor α 2,3 Gal.,
dan SA α 2,6 Gal., atas berlaku substitusi
asam amino Leu 129 Val dengan Ala 134 Val. Mutasi
tersebut dapat melantarkan kemantapan ikatan
SA α 2,6 Gal., atas RBC di hal yang
optimal atas terikat di konformasi cis.
Analisis tanaman kekerabatan yang dibentuk
berdasarkan adegan gen HA atas nukleotida
posisi 208 cukup 612, membentuk beberapa
kelompok mikrob flu burung (Gambar 2). Virus
flu burung yang diisolasi atas tahun 2003, 2004,
2005, dengan per satu mikrob flu burung

yang diisolasi atas tahun 2006 dengan 2007 yang
diteliti membentuk klaster mikrob tersendiri. Ke
dalam kelompok tersebut berdekatan dengan
klaster mikrob flu burung yang diisolasi atas awal
wabah yang telah diperoleh atas gene bank yang
menunjukkan afinitas genetik dengan virus
A/Duck/Hunan/2005, A/Blackbird/Hunan/2004,
dan mikrob induk. Analisis tersebut bersesuaian
dengan berita Wang et al., (2008) yang
melaporkan bahwa mikrob flu burung yang
diisolasi di Indonesia mempunyai kedekatan
genetik dengan mikrob akar Hunan, Cina Selatan.
Virus flu burung yang diisolasi atas tahun
2006, 2007, dengan 2008 membantuk klaster
tersendiri dengan percabangan yang bertambah jauh.
Dari klaster tersebut terbentuk sub-klaster yang
terdiri arah isolat mikrob A/Layer/Jabar/MHW-
RBS-02/2008, A/Chicken/West Java/2006, virus
flu burung akar bani Adam tahun 2007, dengan virus
A/MD/MTL/Pokja-RBS-15/2007. Salah satu
isolat, ialah A/Layer/Jabar/MHW-RBS-02/2008,
menunjukkan percabangan yang paling jauh.
Kondisi tersebut diperkirakan karena
banyaknya mutasi yang berlaku atas fragmen
RBS atas mikrob flu burung tersebut.

Gambaran tanaman kekerabatan adegan gen
HA yang diperoleh TPR di investigasi ini
tidak membuktikan cermin distribusi tertentu dari
spesies ayam atau ceceran geografis, tetapi
secara am klaster mikrob flu burung tersebut
cenderung berdasar atas tahun pengasingan mikrob flu
burung yang diteliti. Pola ceceran mikrob tersebut
bersesuaian dengan produk pengurangan Wang et al.,
(2008) bahwa cermin percabangan pohon
kekerabatan yang digambarkan tak teramati
adanya mikrob yang berkelompok berdasarkan
spesies dengan ceceran geografis tertentu. Kondisi
tersebut berlainan dengan berita Lam et al.,
(2008) bahwa pengurangan filogenetik gen HA virus
flu burung akar Indonesia membuktikan adanya
kelompok spesies, pertama mikrob flu burung
yang diisolasi dari manusia. Virus flu burung
asal ayam membentuk klaster tersendiri
meskipun tak sedia cermin ceceran di antara
spesies ayam yang diteliti.
SIMPULAN
Berdasarkan data persejajaran dalam
penelitian ini kedapatan bahwa mikrob flu burung
subtipe H5N1 yang diisolasi dari berbagai
unggas sejak tahun 2003 cukup 2008, teramati
beberapa masam amino bena TPR yang bersifat
lestari dengan membuktikan kecenderungan
berikatan dengan reseptor corak ayam SA α 2,3
Galaktosa. Salah satu isolat yang diteliti yaitu
A/Layer/Jabar/MHW-RBS-02/2008 (H5N1)
menunjukkan delesi masam amino kedudukan 129 dan
mutasi masam amino I 151 T, yang pada
umumnya terdapat atas mikrob influenza A
subtipe lain, ialah H1, H2 alias H3. Analisis
pohon kekerabatan tak teramati klaster virus
flu burung yang membuktikan cermin distribusi
tertentu dari spesies ayam atau sebaran
geografis akar mikrob yang diteliti.
SARAN
Untuk memafhumi ampas masam amino pada
receptor binding cavity bidang kiri perlu dilakukan
amplifikasi gen HA mikrob flu burung yang
terdapat TPR atas kedudukan loops 220.

An outbreak of highly pathogenic avian pilek A (H5N1) has recently spread to poultry in 9 Asian countries. H5N1 infections have caused >52 human deaths in Vietnam, Thailand, and Cambodia from January 2004 to April 2005. Genomic analyses of H5N1 isolates from birds and humans showed 2 distinct clades with a nonoverlapping geographic distribution. All the viral genes were of avian influenza origin, which indicates absence of reassortment with human pilek viruses. All human H5N1 isolates tested belonged to a single clade and were resistant to the adamantane drugs but sensitive to neuraminidase inhibitors. Most H5N1 isolates from humans were antigenically homogeneous and distinct from avian viruses circulating before the end of 2003. Some 2005 isolates showed evidence of antigenic drift. An updated nonpathogenic H5N1 reference virus, lacking the polybasic cleavage site in the hemagglutinin gene, was produced by reverse genetics in anticipation of the possible need to vaccinate humans.
Highly Pathogenic Avian Influenza (HPAI) H5N1 mikrob is an ongoing public health and socio-economic challenge, particularly in South East Asia. H5N1 is now endemic in poultry in many countries, and represents a major pandemic threat. Here, we describe the evolution of H5N1 mikrob in South East Asia, the reassortment events leading to high genetic diversity in the region, and factors responsible for mikrob spread. The mikrob has evolved with genetic variations affecting virulence, drug-resistance, and adaptation to new host species. The constant surveillance of these changes is of primary importance in the global efforts of the scientific community.
Highly pathogenic avian pilek H5N1 viruses are circulating in many countries. We recently discovered that these viruses have been transmitted to pigs on multiple occasions in Indonesia. To investigate whether avian H5N1 pilek viruses adapted to mammals through their introduction into pigs, we examined the growth of avian and swine isolates in cell culture and compared their pathogenicity in mice. We found that swine isolates were less virulent to mice than avian isolates, suggesting that the viruses became attenuated during their replication in pigs. Continuous surveillance of H5N1 viruses among pigs is clearly warranted.
H5N1 highly pathogenic avian pilek (HPAI) viruses have seriously affected the Asian poultry industry since their recurrence in 2003. The viruses pose a threat of emergence of a global pandemic pilek through point mutation or reassortment leading to a strain that can effectively transmit among humans. In this study, we present phylogenetic evidences for the interlineage reassortment among H5N1 HPAI viruses isolated from humans, cats, and birds in Indonesia, and identify the potential genetic parents of the reassorted genome segments. Parsimony analyses of viral phylogeography suggest that the reassortant viruses may have originated from greater Jakarta and surroundings, and subsequently spread to other regions in the West Java province. In addition, Bayesian methods were used to elucidate the genetic diversity dynamics of the reassortant strain and one of its genetic parents, which revealed a more rapid initial growth of genetic diversity in the reassortant viruses relative to their genetic parent. These results demonstrate that interlineage exchange of genetic information may play a pivotal role in determining viral genetic diversity in a focal population. Moreover, our study also revealed significantly stronger diversifying selection on the M1 and PB2 genes in the lineages preceding and subsequent to the emergence of the reassortant viruses, respectively. We discuss how the corresponding mutations might drive the adaptation and onward transmission of the newly formed reassortant viruses.
Avian pilek mikrob reassortants containing human pilek mikrob hemagglutinins do not replicate in ducks. Two mutations in the receptor-binding site of a human hemagglutinin at residues 226 and 228 allowed replication in ducks. The mutations resulted in a receptor-binding-site sequence identical to the known avian pilek mikrob sequences.
N-Linked glycosylation usually occurs at the sequon, Asn-X-Ser/Thr. In this sequon, the side chain of the hydroxy amino acid (Ser or Thr) may play a direct catalytic role in the enzymatic transfer of core oligosaccharides to the Asn residue. Using recombinant variants of rabies mikrob glycoprotein (RGP), we examined the influence of the hydroxy amino acid on core glycosylation efficiency. A variant of RGP containing a single Asn-X-Ser sequon at Asn was modified by site-directed mutagenesis to change the sequon to either Asn-X-Cys or Asn-X-Thr. The impact of these changes on core glycosylation efficiency was assessed by expressing the variants in a cell-free transcription/translation/glycosylation system and in transfected tissue culture cells. Substitution of Cys at position 39 blocks glycosylation, whereas substitution of Thr dramatically increases core glycosylation efficiency of Asn in both membrane-anchored and secreted forms of RGP. The substitution of Thr for Ser also dramatically enhances the level of expression and cell surface delivery of RGP when the sequon at Asn is the only sequon in the protein. Novel forms of membrane-anchored and secreted RGP which are fully glycosylated at all three sequons were also generated by substitution of Thr at position 39.
N-Linked glycosylation is a common form of zat putih telur processing that can profoundly affect zat putih telur expression, structure, and function. N-Linked glycosylation generally occurs at the sequon Asn-X-Ser/Thr, where X is any amino acid except Pro. To assess the impact of the X amino acid on core glycosylation, rabies mikrob glycoprotein variants were generated by site-directed mutagenesis with each of the 20 common amino acids substituted at the X position of an Asn-X-Ser sequon. The efficiency of core glycosylation at the sequon in each variant was quantified in a rabbit reticulocyte lysate cell-free translation system supplemented with canine pancreas microsomes. The presence of Pro at the X position completely blocked core glycosylation, whereas Trp, Asp, Chi, and Leu were associated with inefficient core glycosylation. The other variants were more efficiently glycosylated, and several were fully glycosylated. These findings demonstrate that the X amino acid is an important determinant of N-linked core-glycosylation efficiency.
The pilek mikrob A hemagglutinin (HA) is a trimeric glycoprotein that contains 3–9 N-linked glycosylation sequons bagi subunit, depending on the strain. The location of these sites is determined by the nucleotide sequence of the HA gene, and, since the viral genome is replicated by an errorprone RNA polymerase, mutations, which add or remove glycosylation sites, occur at a high frequency. Mutations that are not lethal to the mikrob add to the structural diversity of the mikrob population. Factors that determine the glycosylation of the HA are reviewed herein, as are the effects of host-specific glycosylation on receptor binding, fusion activity, and antigenic properties of the virus. Effects of host-specific glycosylation and selection on virulence and on vaccine efficacy and surveillance are discussed. In addition, inadequacies in our understanding of HA glycosylation and its effects on host range are emphasized.
We mutated the virulent avian pilek mikrob A/turkey/Ontario/7732/66 (H5N9)[Q-R-R-R-K-K-R\G at the hemagglutinin (HA) cleavage site] to create a mutant, R(MO-0), with additional basic residues at this site (Q-R-R-R-R-R-K-K-R\G) by reverse genetics. When tested in chicken embryo fibroblast culture, this mutant showed reduced HA cleavability compared to that of the wild-type virus, but its plaque size was not appreciably altered. Virulence of the R(MO-0) mikrob in chickens was lower than that of the wild-type virus. These findings indicate that addition of excessive basic residues to an optimal recognition sequence for HA cleavage enzymes at the cleavage site is deleterious for HA cleavability. Previously, we showed that a mutant containing the suboptimal HA cleavage site sequence for cleavage enzyme recognition also had reduced HA cleavability and virulence compared to the wild-type virus. We conclude that the data presented here further substantiate our belief that the kelas of HA cleavability correlates with the degree of virulence when all other genetic characteristics are considered equal, irrespective of the mechanisms by which HA cleavability is reduced.
